General information about Lamprene

Lamprene (clofazimine) is an antileprosy drug. It inhibits the growth of bacteria that cause leprosy. Clofazimine is active against pathogens that are resistant to other drugs. It also has anti-inflammatory activity and can be used for the treatment of lepra reactions associated with the treatment with other drugs. The drug is used mainly in the complex therapy.

Lamprene is used to treat leprosy (prevention of secondary resistance to drugs known as the sulfones and leprosy reactions in certain types of leprosy). The drug is effective in the treatment of several forms of leprosy resistant to sulfone drugs, as well as leprosy reactions (e.g., erythema nodosum leprosum). Clofazimine is sometimes used in the treatment of multidrug-resistant pulmonary tuberculosis.

General information about Lamprene dosing and administration

Before you start using the drug, it is necessary to use sensitivity testing and determine susceptibility of bacteria. Clofazimine is often used in combination with rifampicin and diaphenylsulfone.

Treatment of lepromatous leprosy: clofazimine – 300 mg 1 time per month and 50 mg 1 time per day as maintenance therapy, rifampicin – 600 mg 1 time per month, diaphenylsulfone – 100 mg 1 time per day as maintenance therapy. The course of treatment is at least 2 years. Clofazimine dose must be increased to 300 mg per day (not more than 3 months) in case of erythema nodosum.

Therapy in children: clofazimine – 5 mg/kg body weight one time per month and 1 mg/kg once daily as a maintenance therapy, rifampicin – 10 mg/kg 1 time per month, diaphenylsulfone – 1.5 mg/kg one time per day as maintenance therapy.

The capsules should be taken with food or milk.

Use with caution in patients with impaired liver and kidney function. In these cases, as well as in case of long-term use of the drug, your doctor may recommend testing both blood and urine every 3 months.

General information about Lamprene side effects

Lamprene therapy may change your basic skin color to red or dark brown (especially in areas exposed to light, in patients with fair skin), as well as hair, conjunctiva, tears, sweat, sputum, urine, feces. Lamprene therapy often leads to dry skin, ichthyosis, pruritus, photosensitivity (increased sensitivity of the skin to sunlight), rash, acne.

Skin: very often – ichthyosis, dry skin; often – itching, redness of the skin; rarely – skin rash, photodermatosis, Lyell’s syndrome.

Digestive system: very often – nausea, vomiting, prolonged diarrhea, discoloration of feces; rarely – anorexia; very – intestinal obstruction, constipation, jaundice, hepatitis, bleeding in the gastrointestinal tract.

Nervous system: rarely – headache and dizziness; very rarely – drowsiness and depression.

Other side effects: very often – loss of body weight; very rarely – fever, lymphadenopathy, anemia.

Nausea, vomiting, abdominal pain, diarrhea, loss of appetite, weight loss – these side effects are usually observed with concomitant diseases of the gastrointestinal tract, such as amoebiasis (infectious diseases caused by the parasite Entamoeba histolytica and characterized by chronic inflammation of the colon with the formation of ulcers). It is necessary to reduce the daily dose or increase the interval between doses of the drug in case of gastrointestinal disorders. Lepra reactions (rarely observed during treatment with clofazimine) can be eliminated by a temporary increase in the dose.

General information about Lamprene contraindications

Clofazimine is contraindicated in people with frequent abdominal pain and diarrhea. Use this drug in the first trimester of pregnancy only under strict indications.
