Janumet action

Janumet is a combination of two hypoglycemic agents – sitagliptin and metformin. It is used to improve glycemic control in patients with type 2 diabetes.

Sitagliptin increases the levels of incretin hormone (associated with regulation of glucose homeostasis). Incretins are secreted into the intestine. In case of normal or elevated blood glucose concentration, incretins enhance insulin synthesis and reduce glucose production. Incretins do not affect the synthesis of insulin and glucagon secretion in response to hypoglycemia. Sitagliptin helps increase the release of insulin and decrease glucagon secretion.

Metformin reduces the concentration of glucose and helps increase glucose tolerance. What are the most common side effects of Janumet?

Cardio-vascular system (blood, hemostasis): in rare cases – megaloblastic anemia (as a result of malabsorption of vitamin B12 and folic acid).

Janumet dosage and administration

Insulin, sulfonylureas, acarbose, NSAIDs, MAO inhibitors, oxytetracycline, ACE inhibitors, clofibrate derivatives, cyclophosphamide, beta-blockers may increase the hypoglycemic effect of Janumet.

Furosemide, amiloride, digoxin, morphine, procainamide, quinidine, quinine, ranitidine, triamterene and vancomycin may increase metformin Cmax by 60%.

Cimetidine slows the elimination of metformin and increases the risk of lactic acidosis.
